Overview

Nuclear receptor coactivator 4 pseudogene 4 (NCOA4P4) is a predicted pseudogene, named to reflect its homology to the functional nuclear receptor coactivator 4 (NCOA4). Pseudogenes are genomic DNA sequences similar to genes but generally nonfunctional either because they are not transcribed/translated, or do not encode a functional protein. NCOA4 itself is an androgen receptor coactivator, but there is no confirmed evidence of biological activity, protein product, or disease association for NCOA4P4. Pseudogenes can sometimes regulate parental gene activity through RNA-based mechanisms, but there is no published data for NCOA4P4 in this context. The NCOA4 gene is functional and encodes a coactivator of several nuclear receptors, with roles in transcription regulation and iron metabolism. It is considered a therapeutic target in certain cancers. Multiple pseudogenes exist for NCOA4 (identified as NCOA4P1, etc.), but NCOA4P4 is not recognized in major databases or the literature, and appears to have no known function or clinical relevance. Pseudogenes can have regulatory effects in some cases, but only select pseudogenes (e.g. within OCT4, PTEN families) are reported to impact disease biology. No such role is documented for NCOA4P4.
