Overview

Nuclear receptor coactivator 6 (NCOA6) is a large, multifunctional transcriptional coactivator protein that interacts with various nuclear hormone receptors (including steroid, thyroid, retinoid, and vitamin D receptors) to enhance their transcriptional activity[5][6][4]. NCOA6 also functions as a general transcriptional coactivator by associating with basal transcription factors and chromatin-modifying enzymes, such as histone acetyltransferases and methyltransferases, thereby influencing epigenetic regulation[3][4][5]. It is involved in a broad range of biological processes, including embryonic development, metabolism, cell proliferation, and the regulation of inflammation via participation in the activation of the NLRP3 inflammasome[1][3][7]. The gene encoding NCOA6 is often amplified and overexpressed in several cancer types, implicating it as an oncogenic driver or facilitator[3]. Multiple isoforms of NCOA6 exist due to alternative splicing, resulting in differential subcellular localization and functional diversity among tissues[7]. Knockouts in mice demonstrate that NCOA6 is essential for viability and the development of several organ systems[3].
