Overview

Nuclear receptor coactivator 7 (NCOA7) is a transcription coactivator that binds to ligand-activated nuclear receptors such as estrogen receptors (ERα, ERβ) and aryl hydrocarbon receptor (AhR), facilitating the recruitment of transcriptional machinery and enhancing gene transcription. NCOA7 also interacts with metabolites (e.g., 3-hydroxyanthranilic acid) to promote immunoregulatory activity, particularly in dendritic cells, through AhR signaling. Tissue expression is highly specific, being prominent in the brain, immune cells (notably conventional dendritic cells), breast, and various tumors. NCOA7 plays roles in hormone response, DNA oxidative damage protection, and immune regulation, and has been linked to cancer prognosis, neurodevelopmental disorders, and immunoregulatory mechanisms. NCOA7 is not a classical therapeutic target but is significant in the regulation and modulation of nuclear receptor-mediated pathways and immunological responses[1][2][3].
