The Nuclear receptor corepressor (NCoR) complex is a multi-protein assembly that serves as a central node in transcriptional repression and epigenetic silencing. It is primarily composed of the scaffold proteins NCOR1 or NCOR2 (also known as SMRT), which recruit and activate the enzymatic subunit Histone Deacetylase 3 (HDAC3), along with accessory proteins such as TBL1, TBLR1, and GPS2 [UniProt, Watson et al., 2012, Nature]. The complex functions by binding to unliganded nuclear receptors—including the thyroid hormone and retinoic acid receptors—and other site-specific transcription factors, leading to the deacetylation of histone tails and the formation of repressive chromatin [Glass and Rosenfeld, 2000, Genes & Development]. In various malignancies, such as acute myeloid leukemia and breast cancer, the NCoR complex is often dysregulated or recruited by oncogenic fusion proteins to suppress genes essential for differentiation and growth control [Lin et al., 1998, Nature; Wong et al., 2001, Molecular and Cellular Biology]. Pharmacological intervention typically involves the use of HDAC inhibitors to counteract the complex's repressive activity or the use of selective nuclear receptor modulators that influence the recruitment of the complex to specific genomic loci [Ghisletti et al., 2009, Molecular Cell]. Beyond its role in oncology, the NCoR complex is a critical regulator of metabolic pathways in the liver and adipose tissue, making it a significant target for research into type 2 diabetes and metabolic syndrome [Li et al., 2011, Cell; Mottis et al., 2013, FEBS Letters].
The NCoR complex acts as a scaffold that recruits and activates Histone Deacetylase 3 (HDAC3) to the promoter regions of target genes, leading to histone deacetylation and transcriptional silencing.
