Nuclear receptor subfamily 1 group D member 1 (NR1D1), also known as REV-ERBα, is a ligand-sensitive transcriptional repressor and a critical component of the circadian clock machinery. It regulates circadian rhythms, metabolic processes, and immune responses by binding to RORE sites on DNA and recruiting corepressor complexes. Heme is an endogenous ligand that stabilizes the interaction with corepressors. NR1D1 is a promising drug target for various diseases, including sleep disorders, metabolic syndrome, inflammatory diseases, cancer, and organ injury-related conditions, owing to its broad regulatory effects on autophagy, immunity, inflammation, metabolism, and aging.
Binds to RORE sites as a monomer or homodimer and recruits corepressor complexes such as NCOR and HDAC3, leading to histone deacetylation and transcriptional repression. Heme binding enhances corepressor interaction.
