Nuclear RNA export factor 3 (NXF3) is a member of the nuclear RNA export factor family involved in mediating the export of cellular mRNA from the nucleus to the cytoplasm for translation[1][2][3]. It contains domains typical of the family (noncanonical RNA-binding, leucine-rich repeats, NTF2-like domain for dimerization, and ubiquitin-associated domain for nucleoporin interaction), though it is unique in having a shortened LRR and ubiquitin-associated domains and an RNA-binding domain that does not bind RNA directly[2][3]. NXF3 is primarily located in the nucleoplasm and is not associated with the nuclear envelope or nucleolus[3]. In humans, it is found to be highly expressed in the testes and regulates specific mRNA export through a Crm1-dependent pathway[1]. Recent studies link NXF3 overexpression to tumor progression in hepatocellular carcinoma, making it a promising prognostic biomarker and a potential target for cancer therapeutics[1][2]. In Drosophila, Nxf3 is essential for piRNA precursor export, suggesting a broader role in RNA metabolism and germline development[4]. However, only limited evidence exists for drugs directly targeting NXF3, with most research focusing on its export mechanism and biological associations.
