Nuclear RNA export factor 3 (NXF3)

Target
NXF3
Molecular classification
mRNA export factor, RNA-binding protein (although NXF3's RNA-binding is atypical and may not bind RNA directly), Adapter for Crm1-dependent nuclear RNA export, Other (belongs to nuclear RNA export factor family)
01

Overview

Nuclear RNA export factor 3 (NXF3) is a member of the nuclear RNA export factor family involved in mediating the export of cellular mRNA from the nucleus to the cytoplasm for translation[1][2][3]. It contains domains typical of the family (noncanonical RNA-binding, leucine-rich repeats, NTF2-like domain for dimerization, and ubiquitin-associated domain for nucleoporin interaction), though it is unique in having a shortened LRR and ubiquitin-associated domains and an RNA-binding domain that does not bind RNA directly[2][3]. NXF3 is primarily located in the nucleoplasm and is not associated with the nuclear envelope or nucleolus[3]. In humans, it is found to be highly expressed in the testes and regulates specific mRNA export through a Crm1-dependent pathway[1]. Recent studies link NXF3 overexpression to tumor progression in hepatocellular carcinoma, making it a promising prognostic biomarker and a potential target for cancer therapeutics[1][2]. In Drosophila, Nxf3 is essential for piRNA precursor export, suggesting a broader role in RNA metabolism and germline development[4]. However, only limited evidence exists for drugs directly targeting NXF3, with most research focusing on its export mechanism and biological associations.

03

Biological functions

Nuclear export of mRNAs from nucleus to cytoplasmTissue-specific expression (notably high in testes)Adapter for Crm1-dependent nuclear export mechanismRegulation of piRNA precursor export (shown in Drosophila, likely analogous roles in mammals)Modulation of TGF-β3 mRNA and protein secretion in Sertoli cellsPotential involvement in tumor cell transformation via dysregulated nucleocytoplasmic transport
04

Disease associations

05

Safety considerations

Currently, no specific safety concerns or therapeutic challenges for direct targeting of NXF3 have been described in clinical settingsGeneral challenges might apply for modulation of mRNA export with off-target effects on cellular homeostasis
06

Interacting drugs

Biomarkers

NXF3 expression level in tumor tissue (especially hepatocellular carcinoma) serves as a prognostic biomarker, correlated with poor survival and higher recurrence rates
