Overview

Nuclear transcription factor Y subunit gamma antisense RNA 1 (NFYC-AS1) is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the NFYC gene, which encodes the gamma subunit of the trimeric NF-Y transcription factor. It is primarily localized in the nucleus and functions as a critical regulator of the cell cycle, particularly the G2/M phase, and promotes cell proliferation and survival (Gatti et al., 2024). NFYC-AS1 is significantly overexpressed in various malignancies, including lung, gastric, and colorectal cancers, and its expression is strongly associated with RB1-mutated tumors and poor patient prognosis (Song et al., 2021). Mechanistically, it acts as a competing endogenous RNA (miRNA sponge) and modulates key oncogenic pathways, such as the MET/c-Myc axis and BIRC6-mediated autophagy (Tong et al., 2022). While no drugs targeting NFYC-AS1 are currently approved for clinical use, preclinical studies utilizing antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) have demonstrated its potential as a therapeutic target by effectively inhibiting tumor growth and inducing apoptosis (ASH, 2024).
