Nuclear transition protein 2 (TNP2, also known as TP2) is a basic nuclear protein expressed in male germ cells, encoded by the human TNP2 gene on chromosome 16[1][7]. It acts during spermatogenesis, specifically in the transitional phase between histone removal and protamine incorporation. TNP2 mediates the exchange of chromosomal proteins by temporarily associating with DNA after histones have been displaced and before protamines are deposited, facilitating proper chromatin compaction required for mature sperm formation. TNP2 activity, along with related transition proteins and specific histone variants, is crucial for sperm DNA condensation and the male germ cell chromatin remodeling process[5][7]. Dysfunction or absence of TNP2 in animal models leads to abnormal sperm chromatin packaging and male infertility, but no therapeutic drugs or clinical biomarkers are currently known[5][7]. TNP2 is classified neither as a receptor, enzyme, transporter, nor a canonical drug target, but rather as a crucial chromatin-associated protein specific to testicular germ cells[7].
