Overview

Nucleic acid-binding protein 1 (NABP1) is a nuclear single-stranded DNA-binding protein that contains a conserved oligonucleotide/oligosaccharide-binding (OB) fold[1][3]. NABP1 is a component of the SOSS (Sensor of single-strand DNA) complex, which functions downstream of the MRN complex to promote the repair of DNA double-strand breaks and regulate the G2/M cell cycle checkpoint[3][4]. NABP1 binds preferentially to single-stranded DNA, especially polypyrimidine sequences, and also interacts with single-stranded RNA[1]. It participates in homologous recombination-dependent DNA repair, nucleotide excision repair, and the activation of ATM kinase signaling pathways following DNA damage[1][2][4]. NABP1's expression is regulated by retinoid-related orphan receptor γ2 (RORγ2) and is highly expressed in rapidly dividing thymocytes, suggesting a role in adaptive immunity and genome rearrangement[1]. Deficiency or improper regulation of NABP1 can impair DNA repair and genomic stability, contributing to cancer susceptibility, with a noted association to acute promyelocytic leukemia[3]. NABP1 operates within high molecular-mass nuclear protein complexes and is essential for cellular responses to genotoxic stresses[1][2][4].
