Overview

This target encompasses the therapeutic cross-linking of DNA/RNA within pathogens and leukocytes contained in blood products, using either chemical agents (S-303, PEN110), photoactive compounds (amotosalen, riboflavin), or similar molecules. Agents intercalate into helical regions of the nucleic acids then induce covalent cross-links—by bis-alkylation (S-303), photo-activation (amotosalen), or ethylenimine chemistry (PEN110)—preventing nucleic acid replication and rendering pathogens and leukocytes biologically inert. Such technologies are applied to improve transfusion safety by preventing transmission of infections and minimizing immune-mediated complications like GVHD. The concept is not a classical protein/enzyme target, but the selective engagement and destruction of nucleic acid in relevant cells and microorganisms.
