Nucleolar and coiled-body phosphoprotein 1 (NOLC1) is a nucleolar protein, also known as NOPP140, that serves as a scaffolding and chaperone molecule in the nucleolus and coiled bodies. It regulates ribosome biogenesis and rRNA transcription by interacting with RNA polymerase I and associated ribosomal processing enzymes. NOLC1 is critical for nucleolus organization, ribosomal RNA processing, and the maintenance of the fibrillar center and dense fibrillar component. It shuttles between the nucleolus and cytoplasm and is involved in several biological processes, including neural crest cell development and cell proliferation regulation. Overexpression or abnormal expression of NOLC1 impacts nucleolar structure and function, represses cell proliferation, and promotes cellular senescence. NOLC1 has documented disease associations in multiple cancer types, with its expression correlating with prognosis in breast cancer and playing roles in tumorigenesis, cellular aging, and, potentially, viral infection.
