Nucleolar complex-associated protein 4 homolog (NOC4L) is a highly conserved protein localized in the nucleolus and nucleoplasm, where it enables RNA binding activity and is essential for ribosome biogenesis, specifically 40S ribosomal subunit assembly and rRNA processing[2][3][4][7][8][9]. NOC4L is homologous to yeast Noc4p and participates in assembling and transporting the 40S ribosomal subunit[3]. It is highly expressed in the testis, adipose tissue, and immune organs, and is dynamically regulated in immune cells such as activated T cells and macrophages[1][3]. In immune contexts, NOC4L regulates T cell activation, proliferation, and post-transcriptional translational control[1]. In macrophages, NOC4L modulates inflammatory pathways and insulin resistance, partly by interacting with the TLR4/TRIF signaling axis; its deficiency leads to enhanced inflammation and metabolic dysfunction in mice[3]. Diseases associated with mutations in this gene include Bowen-Conradi syndrome[4]. There are no known drugs directly targeting NOC4L, and it is not currently considered a major therapeutic target (e.g. receptor, enzyme, transporter) in clinical medicine.
