Overview

Nucleolar protein 11 (NOL11) is a metazoan-specific, nucleolus-localized protein that plays a critical role in the early stages of ribosome biogenesis. It is incorporated into the small subunit (SSU) processome, interacting with proteins such as hUTP4/Cirhin, and is required for both optimal pre-rRNA processing and transcription. NOL11 is essential for maintaining proper nucleolar morphology, and its depletion in cells leads to reduced 18S rRNA maturation, impaired rDNA transcription, and nucleolar stress responses, including p53 stabilization and apoptotic phenotypes in developing organisms. There are no drugs directly targeting NOL11, and it is not considered a classic therapeutic target like enzymes or receptors.
