Overview

Nucleolar protein 12 (NOL12) is a multifunctional RNA-binding protein primarily localized in the nucleolus, nucleus, and cytoplasm of human cells[1][2][3][4]. It plays a critical role in ribosome biogenesis by facilitating the processing and maturation of ribosomal RNA, notably separating large and small ribosomal subunit precursors[1][2][3][4]. NOL12 is also involved in maintaining nucleolar structure and homeostasis through regulation of fibrillarin and nucleolin protein levels[1][4]. Beyond its nucleolar functions, NOL12 is required for maintaining genome integrity, participating in the resolution of DNA stress, and regulating cell cycle progression—deficiency results in p53 activation, G2 arrest, and cellular senescence[1][2][4]. NOL12’s interactome is diverse, including RNA/DNA helicases and DNA repair proteins, which supports a role at the intersection of RNA metabolism and the DNA damage response[2][3]. While NOL12 is not currently classified as a therapeutic target, its functions link it to cellular proliferation, aging, and disorders related to impaired ribosome biogenesis and genome maintenance[1][2][7]. There are no approved drugs or well-established biomarkers related to NOL12, and no direct safety or therapeutic concerns have been described in the literature to date[1][2][3][4].
