Overview

Nucleolar protein 6 (NOL6) is a highly conserved nucleolar RNA-associated protein coded by the NOL6 gene and involved primarily in ribosome biogenesis. It interacts with pre-rRNA primary transcripts and is essential for the formation and function of ribosomes in the nucleolus[2][3][7]. NOL6 is RNA-dependent for its nucleolar localization and is found in at least two isoforms due to alternative splicing[3][7]. Emerging evidence identifies NOL6 as a cancer-associated gene, functioning as an oncogenic driver that promotes cell proliferation and migration and inhibits apoptosis, notably in liver, prostate, and endometrial cancers, partly via the regulation of transcription factors such as TWIST1[2][5][6]. NOL6 is being explored as a potential prognostic biomarker and therapeutic target in oncology[2][5].
