Overview

Nucleoprotein–DNA complexes are macromolecular assemblies consisting of nucleic acids non-covalently or covalently bound to proteins, playing a fundamental role in the organization and regulation of the genome. In a cellular context, these complexes include chromatin, where DNA is wrapped around histone proteins to regulate gene expression and DNA replication. In virology, nucleoprotein-DNA complexes form the core of various DNA viruses, such as Hepatitis B, where the nucleocapsid protein protects the viral genome and facilitates its transport to the host nucleus. These complexes are critical therapeutic targets; for instance, disrupting viral nucleocapsid assembly can halt viral replication, while certain chemotherapeutic agents work by inducing irreversible DNA-protein crosslinks that trigger apoptosis in cancer cells. Furthermore, these complexes are significant in autoimmune pathology, particularly in systemic lupus erythematosus, where they serve as primary antigens for the production of pathogenic autoantibodies.
