Nucleoredoxin (NXN) is a thiol-oxidoreductase enzyme of the thioredoxin family ubiquitously expressed in mammalian cells, shuttling between the cytosol and nucleus. Its redox-dependent interactions regulate signaling pathways such as WNT/β-catenin (via disheveled/DVL), Toll-like receptor 4/MYD88/NF-κB, and protein phosphatase 2A (PP2A). NXN participates in critical processes including cell cycle control, cell proliferation, neuronal development, glycolysis, cytoskeletal regulation, immune responses, and organogenesis. Dysregulation or genetic variants of NXN are implicated in cancer, metabolic and neurodegenerative diseases, and other systemic pathologies. As a central hub for cellular redox signaling, NXN is an important candidate for therapeutic targeting and fundamental cell biology research
