Overview

Nucleoredoxin-like protein 1 (Nxnl1, also known as rod-derived cone viability factor or RdCVF) is a secreted trophic factor with oxidoreductase activity that is primarily expressed in the retina. It is essential for the long-term survival and function of cone photoreceptors by protecting them from oxidative stress and by promoting glucose uptake via a receptor complex involving GLUT1 and BSG proteins[1][5][7][8]. Loss of Nxnl1 expression leads to cone degeneration and is implicated in retinal diseases such as retinitis pigmentosa. While it belongs to the thioredoxin superfamily, Nxnl1 has both enzymatic and signaling roles, with unique properties distinct from canonical nucleoredoxins. There is ongoing research into its therapeutic potential for neurodegenerative diseases of the eye.
