Nucleosome assembly protein 1-like 4 (NAP1L4) is a protein-coding gene and a member of the nucleosome assembly protein (NAP) family, primarily functioning as a histone chaperone in nucleosome assembly[1][2][3][5][10]. NAP1L4 interacts with both core and linker histones, shuttling between the cytoplasm and nucleus to facilitate the transfer and assembly of histones onto DNA, thereby modulating chromatin organization and gene expression[1][2][10]. It is located near the imprinted gene domain at chromosome 11p15.5, an important tumor suppressor locus; changes in this region have been linked to Beckwith-Wiedemann syndrome, Wilms tumor, rhabdomyosarcoma, adrenocortical carcinoma, as well as multiple cancers including lung, ovarian, and breast cancers[1][2][3]. There is no current evidence that NAP1L4 is a direct therapeutic target or is targeted by approved drugs[1][2][10].
