Overview

Nucleotide-binding oligomerization domain-containing protein 2 (NOD2) is an intracellular pattern recognition receptor that detects bacterial peptidoglycan fragments, specifically muramyl dipeptide (MDP). Upon MDP binding, NOD2 oligomerizes, recruits RIPK2, and activates downstream pathways like NF-kappaB and MAPKs, leading to pro-inflammatory cytokine production. Mutations in NOD2 are associated with Crohn's disease and other inflammatory conditions, making it a therapeutic target for modulating the innate immune response.
