Overview

Nucleus accumbens-associated protein 1 (NACC1) is a member of the BTB/POZ and BEN domain protein families, primarily functioning as a transcriptional repressor and coregulator in the nucleus. It regulates multiple cellular processes, including stem cell self-renewal, pluripotency, apoptosis, cell proliferation, and response to innate immunity. NACC1 is frequently overexpressed in a variety of carcinomas, where it promotes tumor progression, cell survival, migration, and chemoresistance by silencing tumor suppressor pathways and modulating transcriptional networks. In addition to oncogenesis, NACC1 mutations are implicated in neurodevelopmental disorders that present with epilepsy and intellectual disabilities, highlighting its importance for normal neural development. The protein forms homo- and heterodimers through its BTB/POZ and BEN domains, facilitating DNA binding and the recruitment of repressive chromatin complexes. While direct pharmacological modulators are not yet available, NACC1 represents a promising target for future cancer therapies and precision medicine approaches in neurodevelopmental disorders.
