Nudix hydrolase 22 (NUDT22) is a member of the Nudix (nucleoside diphosphate linked moiety X) protein family and functions as a Mg²⁺-dependent UDP-glucose and UDP-galactose hydrolase, hydrolyzing these substrates into UMP and glucose 1-phosphate or galactose 1-phosphate, respectively[1][2]. It possesses a unique structural fold within the Nudix family, including a modified NUDIX box and a distinct N-terminal domain associated with substrate recognition[1]. Located in the nucleoplasm, NUDT22 is involved in nucleotide and carbohydrate metabolism and may play roles in cellular metabolic homeostasis[2][3]. Expression analyses indicate potential relevance in cancer, as levels can differ between normal and tumor tissues[3]. Currently, there are no known drugs that target NUDT22, nor is it established as a biomarker or has documented safety concerns for therapeutic intervention[2][3].
