NUT family member 2B is a nuclear protein that, by itself, is uncharacterized and part of an understudied gene family. However, when fused to the 14-3-3 protein YWHAE (via chromosomal translocation), it creates an oncogenic fusion protein strongly implicated in aggressive, high-grade endometrial stromal sarcoma (ESS), clear cell sarcoma of the kidney (CCSK), and other soft tissue tumors. The YWHAE-NUTM2B fusion enhances cell proliferation and alters cell cycle regulation by dysregulating key signaling pathways (RAF/MEK/MAPK and Hippo/YAP-TAZ). This fusion is now recognized as an important biomarker for diagnosis and prognosis in several rare pediatric and adult cancers. There is currently no direct therapeutic agent targeting NUT family member 2B or its protein product; its clinical relevance is almost exclusively through oncogenic gene fusions, which are often used for tumor classification and prognosis.
