Overview

Nutritional balance refers to the physiological state achieved through optimal intake and metabolism of macronutrients (carbohydrates, proteins, fats), micronutrients (vitamins, minerals), and other dietary components to maintain metabolic homeostasis and support cellular functions.[1][5] It underpins key processes like one-carbon metabolism, which provides methyl donors such as S-adenosylmethionine (SAM) for DNA and histone methylation, and influences epigenetic regulation via metabolites like α-ketoglutarate, acetyl-CoA, and NAD+ that modulate enzymes such as TET proteins, HATs, HDACs, and sirtuins.[1] Disruptions in nutritional balance, such as deficiencies in folate, methionine, choline, or vitamins B6/B12, impair these pathways, leading to aberrant gene expression, oxidative stress, DNA damage, and heightened disease risk including cancer and metabolic disorders.[1][2] In disease contexts, poor nutritional balance contributes to pathological states like hepatocarcinoma from methyl deficiency or metabolic syndrome through altered insulin signaling and lipid metabolism.[1][2] While not a specific molecular entity amenable to direct drug targeting, achieving nutritional balance via diet prevents epigenomic instability and supports overall health, with antioxidants and bioactive compounds like polyphenols aiding in maintaining metabolic and epigenetic equilibrium.[1]
