The NY-ESO-1 peptide–HLA class II complex is a molecular assembly consisting of an epitope derived from the New York esophageal squamous cell carcinoma 1 (NY-ESO-1) protein bound to a Human Leukocyte Antigen (HLA) class II molecule. NY-ESO-1 is a prominent cancer-testis antigen (CTAG1B) that is highly expressed in various solid tumors, such as melanoma and synovial sarcoma, but is restricted to the testis in healthy adults, making it an ideal target for immunotherapy (UniProt P78358). On dendritic cells, this complex serves as the primary signal for the activation and expansion of CD4+ T helper cells, which are essential for coordinating long-term immune memory and supporting CD8+ cytotoxic T-cell functions (PMID: 11160657). Therapeutic interventions targeting this complex include viral vector vaccines like LV305, which are designed to induce the expression and presentation of NY-ESO-1 by dendritic cells in vivo (PMID: 30559156). Additionally, adoptive T-cell therapies utilizing TCRs specific for NY-ESO-1/HLA-II complexes are being developed to provide direct anti-tumor activity. The specificity of NY-ESO-1 expression minimizes the risk of systemic toxicity, although challenges such as HLA downregulation and the immunosuppressive tumor microenvironment remain significant hurdles.
