The **NY-ESO-1 peptide (SLLMWITQC; residues 157–165) presented by HLA-A*02:01** is an immunodominant epitope derived from NY-ESO-1, a prototypic cancer-testis antigen highly expressed in several solid and hematological malignancies but absent from essential normal tissues except germ and placental cells[1][4][6]. The HLA-A*02:01/NY-ESO-1 complex is recognized by cytotoxic T lymphocytes, enabling targeted immune destruction of cancer cells[2][3][4][5]. This specificity makes it a central focus of adoptive T cell therapies, therapeutic cancer vaccines, and biomarker-driven immunotherapy trials, offering documented clinical responses in melanoma, synovial sarcoma, neuroblastoma, and multiple myeloma[3][4][5][6]. The main therapeutic challenges are tumor immune escape through antigen or MHC loss and limited eligibility because of HLA restriction, but safety has been excellent with little risk of autoimmunity due to restricted antigen expression[4][5][6].
