The NY-ESO-1 peptide/MHC complex consists of a short peptide derived from the cancer-testis antigen NY-ESO-1 presented by an MHC class I molecule (such as HLA-A*02:01) on the cell surface. This composite structure is specifically recognized by the T cell receptor of cytotoxic T lymphocytes and can induce robust anti-tumor immune responses[4][5][6]. NY-ESO-1 pMHC complexes are among the most immunogenic tumor antigens yet identified, and are used as targets in adoptive T cell therapy, cancer vaccines, and bispecific antibodies. They serve as a biomarker for selecting patients who might benefit from targeted immunotherapies. Therapeutic engagement centers on recognition by engineered TCR or BiTE molecules, facilitating the targeted elimination of NY-ESO-1–expressing tumor cells[1][3][5].
