NY-ESO-1 peptides presented by HLA-DRA + HLA-DRB3*02:02 constitute a specific peptide-major histocompatibility complex (pMHC) class II target for cancer immunotherapy. NY-ESO-1, also known as Cancer-Testis Antigen 1B (CTAG1B), is a highly immunogenic protein expressed in various cancers, including melanoma, synovial sarcoma, and lung cancer, while remaining restricted to germ cells in healthy tissues (Source 1.1.2, 1.2.5). The HLA-DRB3*02:02 allele is a prevalent MHC class II molecule, found in approximately 50% of the Caucasian population, and is responsible for presenting the immunodominant NY-ESO-1(123–137) peptide epitope (Source 1.2.1). Recognition of this pMHC complex by CD4+ T cells induces a potent Th1-polarized immune response and direct cytotoxic activity against tumor cells (Source 1.2.1, 1.2.3). Therapeutic approaches targeting this complex primarily involve adoptive T-cell transfer (ACT) using T-cell receptors (TCRs) engineered to recognize the NY-ESO-1/HLA-DRB3*02:02 complex (Source 1.2.5). These engineered CD4+ TCR-T cells have shown significant anti-tumor efficacy in preclinical models and are being advanced into clinical trials for both adult and pediatric patients (Source 1.2.2, 1.2.5). This target is particularly valuable for expanding the reach of immunotherapy to patients who do not express the more common HLA-A*02:01 allele (Source 1.2.5).
