Overview

O^6-alkylguanine-DNA alkyltransferase (AGT), also known as O^6-methylguanine-DNA methyltransferase (MGMT), is a DNA repair enzyme that removes alkyl groups from the O^6 position of guanine in DNA. The SNAP-tag is a self-labeling protein tag derived from AGT that can be genetically fused to any protein of interest, enabling site-specific covalent labeling with synthetic molecules. It is used in antibody-directed targeting strategies and has been explored in cancer therapy and adoptive cell therapies.

Mechanism of action

The SNAP-tag reacts specifically and covalently with substrates containing an O^6-benzylguanine (BG) moiety. Upon reaction, the tag forms a stable thioether bond between a reactive cysteine residue in the tag and the BG derivative.
