Overview

O6-alkylguanine-DNA alkyltransferase (AGT), also known as O6-methylguanine-DNA methyltransferase (MGMT), is a DNA repair protein that removes alkyl groups from the O6 position of guanine in DNA. This direct reversal mechanism protects cells from the mutagenic, cytotoxic, and carcinogenic effects of alkylating agents. The enzyme acts through a "suicide" mechanism, where it is irreversibly inactivated after accepting an alkyl group. High levels of AGT can confer resistance to chemotherapy drugs, making it a target for therapeutic intervention.
