Overview

OBSCN antisense RNA 1 (OBSCN-AS1) is a nuclear long non-coding RNA (lncRNA) gene located on the antisense strand of the OBSCN gene, partially overlapping with coding regions at the 5' end. It produces two transcript variants and is primarily found in the nucleus. OBSCN-AS1 plays a direct regulatory role in activating OBSCN transcription, likely through modulating chromatin structure (notably by promoting H3K4 trimethylation and recruiting RNA polymerase II). Downregulation of OBSCN-AS1 in breast cancer, especially triple-negative breast cancer, is associated with decreased OBSCN expression, increased migratory and invasive cell behavior, and higher metastatic potential. CRISPR-based activation of OBSCN-AS1 can restore OBSCN levels, suppressing migration and metastasis, suggesting its value as both a prognostic biomarker and a candidate therapeutic target for metastatic breast cancer. No drugs are currently known to target OBSCN-AS1 directly, but antisense oligonucleotide technology and CRISPR tools have been utilized in research to modulate its activity[1][2][3][4][5].
