Overview

OCIA domain-containing protein 1 (OCIAD1) is an integral membrane protein predominantly localized to the inner mitochondrial membrane, with two predicted transmembrane domains[1][3]. It acts as a chaperone and assembly factor specifically for mitochondrial respiratory Complex III, facilitating the correct processing of the CYC1 subunit and interacting with prohibitin scaffolding complexes[1]. Although OCIAD1 is annotated as a mitochondrial protein, previous reports also suggest localization to endosomes and peroxisomes[1]. OCIAD1 maintains normal mitochondrial function; its knockdown results in isolated deficiencies in Complex III assembly, potentially contributing to mitochondrial diseases[1]. OCIAD1 is also a cellular substrate of the hepatitis C virus NS3-4A protease, implicating it in viral infection-related mitochondrial dysfunction[3]. OCIAD1 has additional roles proposed in stem cell biology and has been reported as an ovarian cancer immunoreactive antigen, although its precise role in cancer remains incompletely defined[1][2][3]. There are currently no drugs known to directly target OCIAD1, and no established drug-based mechanisms or specific safety concerns have been described.
