Overview

Odontoblast-like cell differentiation is a complex biological process where dental mesenchymal progenitor cells, such as dental pulp stem cells (DPSCs), differentiate into cells that resemble functional odontoblasts (Gronthos et al., 2000; PMID: 11135448). These cells are characterized by their ability to secrete a collagenous matrix and mineralize it to form dentin, a process known as dentinogenesis (Thesleff et al., 2001; PMID: 11331024). This differentiation is regulated by a variety of signaling molecules, including bone morphogenetic proteins (BMPs), transforming growth factor-beta (TGF-beta), and fibroblast growth factors (FGFs) (Nakashima and Reddi, 2003; PMID: 12679490). In clinical practice, inducing this process is a primary goal of regenerative endodontics and vital pulp therapy, where materials like mineral trioxide aggregate (MTA) are used to stimulate reparative dentin formation (Torabinejad and Parirokh, 2010; PMID: 20113774). Understanding the molecular mechanisms of this differentiation is crucial for developing advanced tissue engineering strategies to treat dental caries and pulp injuries. Key markers of this process include dentin sialophosphoprotein (DSPP) and dentin matrix protein 1 (DMP1), which are essential for proper biomineralization (D'Souza et al., 1997; PMID: 9248310). Therapeutic interventions often aim to modulate the Wnt/beta-catenin or MAPK signaling pathways to enhance the odontogenic potential of stem cells (Liu et al., 2014; PMID: 24906579).
