Overview

Odontoblasts are neural crest-derived, tall columnar cells lining the dental pulp, responsible for the formation of dentin—the mineralized tissue beneath tooth enamel[4][5][1]. They play a critical role in secreting dentin matrix and its mineralization throughout life[3][4][5]. Recent research shows odontoblasts also function as mechanosensory receptor cells: mechanical stimulation of odontoblasts (such as through dentinal fluid movement) activates mechanosensitive ion channels—including Piezo1, TRPV1, TRPV2, TRPV4, and TRPA1—resulting in intracellular calcium signaling and neurotransmitter (ATP) release, which can communicate with sensory neurons to generate pain or hypersensitivity[1][2][4]. Odontoblasts also participate in immune surveillance and defense within the tooth pulp[5]. "Odontoblast stimulation" itself describes activation of these cells (mechanically or chemically) but is not a molecular entity and should not be used as a formal drug or receptor target.
