Odontogenic ameloblast-associated protein (ODAM) is a secreted protein predominantly expressed during tooth development by maturation stage ameloblasts and in the junctional epithelium (JE), where it accumulates at the interface between the basal lamina and enamel[1][2]. ODAM plays a fundamental role in mediating adhesion between epithelial cells and the tooth surface, regulating JE regeneration and epithelial integrity, and participating in cell signaling events related to cytoskeletal reorganization via the integrin-ODAM-ARHGEF5-RhoA pathway. Loss of ODAM expression is associated with pathological states such as periodontal disease and peri-implantitis, while its detection in gingival fluids serves as a biomarker for tissue damage and disease progression. Research has also shown anti-cancer properties of ODAM and its utility as a prognostic marker in malignant transformation, although its precise mechanism in neoplastic tissues remains under investigation[1][2]. Currently, no therapeutic drugs directly target ODAM, but its potential as a target for regeneration therapies is being explored.
