Overview

Odontogenic differentiation is the complex biological process through which undifferentiated mesenchymal stem cells, primarily dental pulp stem cells (DPSCs) or stem cells from the apical papilla (SCAP), transition into specialized odontoblasts. These odontoblasts are uniquely responsible for the synthesis and secretion of the dentin matrix and its subsequent mineralization (NCBI Bookshelf, 'Dentinogenesis'). This process is tightly regulated by various signaling pathways, including the Bone Morphogenetic Protein (BMP), Transforming Growth Factor-beta (TGF-β), and Wnt/β-catenin cascades (PubMed, PMID: 31331405). While not a single molecular target itself, odontogenic differentiation is a central focus in regenerative endodontics and dental tissue engineering. Clinicians and researchers aim to modulate this process using bioactive materials like Mineral Trioxide Aggregate (MTA) to stimulate reparative dentin formation following pulp injury (PubMed, PMID: 29033374). Failures or genetic mutations affecting this differentiation pathway lead to structural dental disorders such as Dentinogenesis Imperfecta and Dentin Dysplasia (StatPearls, 'Dentinogenesis Imperfecta').
