OIP5 antisense RNA 1 (OIP5-AS1), also known as Cyrano, is a highly conserved long non-coding RNA expressed broadly across mammalian tissues, with notable enrichment in the nervous system and muscle. OIP5-AS1 regulates diverse cellular functions, including cell survival, proliferation, apoptosis, and differentiation, primarily through interaction with specific proteins and microRNAs. Its role as a ceRNA enables it to modulate gene expression post-transcriptionally by sequestering RNA-binding proteins (e.g., HuR) or microRNAs (e.g., miR-340-5p, miR-424), thereby affecting downstream gene targets such as ZEB family transcription factors and SIRT1. OIP5-AS1 is implicated in the development and progression of multiple disease states, especially cancers where it promotes cell migration, invasion, and metastasis through regulation of EMT and oncogenic pathways. Its expression profile and regulatory activities make it a candidate biomarker for malignancy and a potential therapeutic target, although no drugs currently target OIP5-AS1 in clinical practice. Further research is needed to clarify its utility and safety as a target for novel RNA-based therapies.
