Overview

Olfactory receptor 2D2 (OR2D2) is a member of the large G protein-coupled receptor (GPCR) family, specifically the olfactory receptor subfamily [1, 2]. Primarily expressed in the olfactory epithelium, it is responsible for detecting odorant molecules and initiating the signal transduction cascade that leads to the perception of smell [2, 4]. Beyond its role in olfaction, OR2D2 is ectopically expressed in various non-olfactory tissues, including the brain, colon, and thyroid gland [2, 4]. Research has linked dysregulation of OR2D2 to neurodegenerative and psychiatric conditions, such as Parkinson's disease and schizophrenia, where its expression is significantly altered in regions like the substantia nigra [3, 4]. Additionally, it is being investigated as a potential biomarker and therapeutic target in certain cancers, such as glioma, where it may influence tumor-related cellular functions [4]. While specific clinical drugs targeting OR2D2 are not yet available, it is known to interact with ligands like androstenone and is a subject of interest for developing novel sensory-based therapies [5, 6].
