Olfactory receptor 8D2 (OR8D2) is a member of the G protein-coupled receptor (GPCR) superfamily, specifically categorized within the Class A rhodopsin-like receptors (UniProt Q9GZM6; GeneCards). Its primary biological function is the detection of volatile odorant molecules in the olfactory epithelium, where it initiates a signal transduction cascade involving the olfactory-specific G protein (Golf) and adenylate cyclase III to facilitate the perception of smell (Wikipedia). Beyond the nasal cavity, OR8D2 exhibits ectopic expression in various non-olfactory tissues such as the colon, heart, and testicles, suggesting roles in non-chemosensory physiological processes (Human Protein Atlas). In clinical research, OR8D2 has gained attention as a component of a 15-gene signature used to predict the risk of recurrence and overall prognosis in patients with colon cancer (Xu et al., 2017). This association highlights its potential utility as a biomarker for cancer progression and a possible therapeutic target for inhibiting tumor growth or metastasis. Currently, there are no approved drugs that specifically target OR8D2, and the identification of its specific endogenous or exogenous ligands remains an area of active investigation. Therapeutic development faces challenges such as the high sequence homology among the hundreds of human olfactory receptors, which complicates the design of selective agonists or antagonists. Despite these hurdles, the ectopic presence of OR8D2 in malignant tissues makes it an intriguing candidate for precision medicine and novel oncology treatments.
