Overview

Olfactory receptor family 10 subfamily A member 6 (OR10A6) is a member of the olfactory receptor (OR) gene family, which encodes G protein-coupled receptors responsible for the detection and transduction of odorant signals in the nose[2][3][4]. OR10A6, like other olfactory receptors, possesses a characteristic 7-transmembrane domain structure. Uniquely, OR10A6 is also expressed in human skin, specifically basal layer keratinocytes, and is implicated in epidermal differentiation and keratinization by regulating calcium and cAMP signaling in response to its agonist, 3PPP[1]. While its main recognized function is odorant detection, emerging evidence suggests distinct tissue-specific roles for this receptor beyond the olfactory system[1][2]. It is not currently implicated as a biomarker or therapeutic target in major diseases, but may represent a new target for modulation of skin barrier function and keratinization[1].

Mechanism of action

Agonism: 3PPP binds to and activates OR10A6, leading to a G protein-coupled signaling cascade that increases intracellular Ca\(^{2+}\), cAMP, and promotes keratinocyte differentiation
