Olfactory receptor family 10 subfamily G member 7 (OR10G7) is a G protein-coupled receptor encoded by a single-exon gene and is a member of the largest gene family in the human genome. While best known for its canonical role in the nose where it binds odorant molecules to initiate neuronal odor perception, this receptor is also ectopically expressed in human skin, particularly in undifferentiated keratinocytes. In keratinocytes, OR10G7 expression is increased in atopic dermatitis and is responsive to specific odorants such as acetophenone and eugenol, triggering ATP production—a process implicated in chemosensory modulation in the skin. Like other olfactory receptors, OR10G7 spans the membrane seven times and utilizes G protein-mediated transduction to convert extracellular chemical (odorant) signals into intracellular responses. OR10G7 is not considered a major therapeutic drug target, but it is a true receptor in molecular and structural terms. There are no widely recognized drug modulators except for odorant ligands (acetophenone, eugenol); no established therapeutic drugs or safety concerns have been reported. There is evidence for a unique role in the skin and atopic dermatitis, but no current biomarker or clinical use is established.
