Overview

Olfactory receptor family 10 subfamily H member 1 (OR10H1) is a member of the largest family of G protein-coupled receptors, responsible for recognizing odorant molecules and initiating neuronal responses in the olfactory system. Unlike most olfactory receptors, OR10H1 has highly specific expression in the urinary bladder, where its abundance substantially increases in bladder cancer tissues. In addition to its canonical olfactory functions, OR10H1 mediates key cellular processes in bladder cancer cells, including cytoskeletal reorganization, modulation of cell viability, proliferation, migration, and induction of cell cycle arrest when stimulated by its specific agonist, Sandranol. It also modulates the secretion of ATP, serotonin, and cytokines (IL10, IL15). OR10H1 is being investigated as a biomarker and therapeutic target for bladder cancer, with its mRNA detectable at higher levels in the urine of affected patients[1][3][4][5].

Mechanism of action

Agonists (Sandranol/Sandalore) bind OR10H1, activating adenylyl cyclase and increasing intracellular cAMP and Ca(2+), with downstream effects of ATP and serotonin secretion, cytoskeletal remodeling, inhibiting cell proliferation/migration, inducing cell cycle arrest (G1), and limited apoptosis. Induces cytokine synthesis (including IL10, IL15), potentially affecting immune response in bladder cancer.
