Overview

Olfactory receptor family 2 subfamily A member 7 (OR2A7) is a member of the largest gene family in the human genome, the olfactory receptor family, which mediates the recognition of odorant molecules in the nasal epithelium and triggers the neuronal signals responsible for the sense of smell. OR2A7, like other olfactory receptors, is a G protein-coupled receptor (GPCR) with a canonical structure of seven transmembrane domains, three intracellular loops, and three extracellular loops. Ligand binding (by small volatile molecules) initiates G protein-mediated signal transduction, typically resulting in activation of adenylate cyclase, increased cAMP, and neuronal depolarization through calcium influx. OR2A7 is not currently a known therapeutic drug target, though it has been studied functionally for its ligand selectivity and ectopic expression, particularly in the colon, where it may have roles in host–microbiome communication. Experimental research has identified compounds that activate this receptor, but no drugs are approved that specifically target OR2A7. No biomarkers or safety concerns are recognized for clinical settings
