Olfactory receptor family 2 subfamily AT member 4 (OR2AT4) is a G protein-coupled receptor primarily known for its role in olfaction but also ectopically expressed in non-nasal tissues such as skin. It recognizes odorant molecules like the synthetic compound sandalore. Upon activation—particularly by sandalore—OR2AT4 triggers intracellular signaling cascades involving calcium influx and MAP kinase phosphorylation. This leads to enhanced proliferation and migration of keratinocytes during wound healing as well as promotion of hair growth through anti-apoptotic effects and upregulation of growth factors such as IGF1. The gene is associated with certain conditions including telogen effluvium and inflammatory bowel disease. Its primary molecular function is signal transduction via GPCR pathways.[1][6][7]
Activation by sandalore leads to G protein-mediated signaling, elevation of intracellular Ca(2+), cAMP-dependent pathways, phosphorylation of MAPK/ERK kinases, promotion of cell proliferation and migration in keratinocytes, decreased apoptosis in hair follicles, increased IGF1 production[1][6][7]
