Olfactory receptor family 2 subfamily T member 4 (OR2T4) is a human G protein-coupled receptor (GPCR) expressed predominantly in the olfactory epithelium, where it participates in the detection of odorant molecules, particularly terpenes, contributing to the sense of smell. Like other olfactory receptors, it possesses a seven-transmembrane domain structure and mediates signal transduction via heterotrimeric G proteins, resulting in neuronal activation and perception of odors. Emerging studies indicate that OR2T4 is also expressed ectopically in non-olfactory tissues and may be involved in additional biological processes and diseases, including anosmia, asthma, and potentially certain cancers, although these disease roles remain under investigation. Experimentally, OR2T4 is activated by several plant-derived compounds, such as pinene and lilial, and its responsiveness to different odorants varies based on genetic polymorphisms[2][3][5][6][7].
Odorant binding triggers G protein-coupled signaling, leading to increased intracellular cAMP and Ca²⁺, and activation of MAPK/ERK kinases[2][5][6]
