Olfactory receptor family 4 subfamily D member 2 (OR4D2) is a member of the large family of olfactory receptor proteins, which are a subset of G protein-coupled receptors (GPCRs) characterized by their seven transmembrane domains. OR4D2 is primarily expressed in nasal olfactory epithelium, where it is responsible for detecting specific odorant molecules, leading to signal transduction via G protein-mediated pathways, and ultimately contributing to the sense of smell. Like other olfactory receptors, OR4D2 is encoded by a single exon gene and operates through ligand-induced conformational changes that enable interaction with intracellular G proteins. Although olfactory receptors such as OR4D2 are predominantly involved in sensory function and not currently established as therapeutic targets in disease contexts, they are critical for chemosensory signal perception and have structural features typical of the broader GPCR superfamily[2][4][5].
Agonists (odorant molecules) bind to the extracellular domain, inducing a conformational change that triggers G protein signaling, leading to neuronal activation and ultimately perception of smell[2][4][5].
