Overview

Olfactory receptor family 5 subfamily K member 3 (OR5K3) is a G protein-coupled receptor (GPCR) predominantly expressed in the olfactory epithelium of the human nose, where it binds endogenous odorant molecules, initiating the neuronal signal cascade responsible for odor perception[6][7][9]. Like other olfactory receptors, OR5K3 has a canonical seven-transmembrane domain structure, interacts with odorants through a diverse binding pocket, and signals via intracellular G proteins to trigger downstream neuronal activity. The OR5K3 gene is one of approximately 400 functional olfactory receptor genes in humans, each mediating detection of a broad set of odorant molecules with some combinatorial overlap. While olfactory receptors as a family have key roles in chemosensation and are structurally related to other GPCR drug targets, OR5K3 itself has no documented therapeutic applications, disease associations, or known drug modulators at present[6][7][9].

Mechanism of action

Agonists (natural odorants) bind to the orthosteric site, induce conformational change, activate intracellular G proteins (mostly *G_olf*), leading to signal transduction via cAMP production[1][7][9]. No specific small-molecule antagonists or agonists of OR5K3 described in published literature.
