Overview

Oligodendrocyte progenitor cell (OPC) differentiation refers to the process by which OPCs mature into myelinating oligodendrocytes, a critical step for proper central nervous system (CNS) function. Mature oligodendrocytes are responsible for forming the myelin sheath that insulates neuronal axons and enables rapid signal transmission. The process involves several stages: proliferation, lineage commitment, premyelinating stage and maturation. Key molecular regulators include transcription factors (OLIG1, OLIG2, SOX10, NKX2.2), chromatin modifiers & miRNAs, extracellular matrix components, and integrins. Proper regulation of this pathway is vital for CNS health; defects lead to demyelinating diseases such as multiple sclerosis. Enhancing endogenous or transplanted OPC differentiation holds therapeutic promise for remyelination strategies after injury or disease.
