Overview

Oligosaccharides are short-chain carbohydrates composed of three to ten monosaccharide units, typically found as part of glycoconjugates like glycoproteins and glycolipids on cell surfaces [1, 7]. They play essential roles in biological processes including cell-cell recognition, the modulation of immune responses, and the stabilization of protein structures [3, 7]. In various disease states, specific oligosaccharide structures act as markers for cancer metastasis, sites for inflammatory cell recruitment, or receptors for viral and bacterial pathogens [3, 8]. Therapeutically, oligosaccharides can serve as both targets and active agents; for instance, monoclonal antibodies target tumor-associated glycans like GD2, while synthetic oligosaccharides like fondaparinux and acarbose are used to treat thrombosis and diabetes, respectively [1, 6, 9]. The immense structural diversity of these molecules, resulting from varied glycosidic linkages and branching, offers a vast but complex landscape for drug discovery and vaccine development [10, 15].

Mechanism of action

Inhibition of carbohydrate-binding proteins (lectins/selectins), induction of antibody-dependent cellular cytotoxicity (ADCC) via glycan-binding antibodies, competitive inhibition of microbial attachment, or modulation of enzyme activity through glycomimicry [1, 3, 4, 12].
