Overview

Trophoblast glycoprotein (TPBG), also known as 5T4 or WAIF1, is a single-pass transmembrane glycoprotein highly expressed in placental trophoblasts during fetal development and overexpressed in a variety of carcinomas. It plays a role in embryonic development, Wnt signaling, cell adhesion and motility, and angiogenesis. TPBG is considered an oncofetal antigen and is associated with poor prognosis in several cancers. It is a target for immunotherapy and antibody-drug conjugates.

Mechanism of action

Monoclonal antibodies such as mAb5T4 target TPBG for antibody-directed immunotherapy; Antibody-drug conjugates (ADCs) use TPBG as a target antigen to deliver cytotoxic drugs selectively to tumor cells; Acts as an antagonist/inhibitor within the canonical Wnt pathway; Downregulation leads to loss of E-cadherin function/cytoskeletal disruption; Orchestrates pericyte movement via modulation of chemokine axes like CXCL12/CXCR4/CXCR7/pERK.
